Misao Amano is Sasami Kawai's best friend, but when she is Pixy Misa, she is her rival.

Misao is notable for appearing in several anime titles in and out of the Tenchi Muyo franchise. In most appearances, she is depicted as a shy and soft-spoken girl who has difficulty making friends with others due to her reserved personality.

Pretty Sammy

In both the OVA and TV anime, Misao is portrayed to be Sasami's best friend in their fourth grade class who is unknowingly hypnotized by Ramiya to transform into Pixy Misa, a magical girl used by Ramia to cause trouble for Pretty Sammy on Earth to jeopardize Tsunami's chances of being the queen of Juraihelm. Unlike the shy and reserved personality of Misao, Misa has an aggressive and lively personality who is prone to performing flashy entrances and demonstrating exaggerated dramatics. Yet like Misao, Misa is physically weak and lacking in stamina. According to Ramiya, Misa represents repressed aspects of Misao's personality.

While putting on a cheerful and quiet persona in front of Sasami, Misao is internally lonely and unhappy thanks to having a family situation where her parents lack time to see her due to having much of their time spent working. She is largely unaware of her identity as Pixy Misa throughout Magical Project S until later in the series and realizing Sasami's true identity as Pretty Sammy. This does drive Misao to be overwhelmed by guilt for her actions until Sasami forgives her. Misa would come to Sammy's aid in the final episodes of the series when confronted with the threat to Earth brought about by Romio.

Battle Programmer Shirase

Here, Misao is the great-niece of the show's protagonist, Akira Shirase. She retains her shy and reserved personality from the Pretty Sammy series, yet is now in the sixth grade. Misao normally visits Akira to cook and tend to him at his apartment out of concern for him and refers to him as "onii-chan", showing a great amount of care for him. She is frequently a victim of the show's running gag where she and Akira are caught in a compromising position whenever a client for the latter arrives to see him at his apartment.

Sasami: Magical Girls Club

Known here as Misao Shinohara, she has a slightly lighter shade of blue for hair color and has a bow in her hair compared to earlier designs of her character. Misao starts off with an introverted personality thanks to her magical ability to create dark worm-like creatures whenever she is scared or under any great deal of stress. Making friends with Sasami, Misao gains confidence in herself and becomes more outgoing as she participates in the activities of the Magical Girls Club. She has a crush on classmate Monta, yet is too shy to confess her feelings to him.

Later in the series, Misao finds herself conflicted over whether or not to live in the magic world when she becomes acquainted with a group of magical folk led by the Chief Sorceress. Misao is manipulated by the Chief Sorceress into turning against the Magical Girls Club, as the latter plots to use Misao as a pawn in her plans to take over the human world.
