Having kept her end of the bargain, Ryuko demands answers from her homeroom teacher, Aikuro Mikisugi, and learns the secret behind the power of Senketsu and the Goku Uniforms. Meanwhile, Satsuki Kiryuin returns home to claim a power of her own.

As a little girl, Satsuki looks forward to wearing Junketsu, her father's wedding gift. During Aikuro's history lesson about Hitler's rise to power, Aikuro hightail out of class with Ryuko following him. Out of nowhere, Mako comes flying like a dart. On her back, it's Aikuro's letter. Afterwards, Aikuro chats with Ryuko in private. Ryuko demands to know how he knew Senketsu and her father. Aikuro explains about Life Fibers and her father's work. He is an acquaintance of Ryuko's father. After receiving Ryuko's father's letter, Aikuro finds the Kamui Uniform and begins his schemes. He tells Ryuko that he will investigate.

In the sewing laboratory, Inumuta and a blond man have the men place threads with level five Life Fiber on a specimen. Suddenly, the specimen transforms into a brutish hulk and tries to attack. Inumuta subdues the specimen. The blond man notes that they cannot make uniforms above level 3 without losing to insanity. It's a miracle that Ryuko could wear a Kamui Uniform that is made of 100% Life Fibers. Over at the Kiryuin Manor, Satsuki barges in to take Junketsu, her Kamui Uniform. She strips her clothes off and slits her index finger to feed her uniform blood. Junketsu awakens and takes over Satsuki. At Mako's home, Barazo, Mataro, and Guts quickly fall asleep. Sukuyo gives Ryuko some pajamas, and she asks Ryuko if she likes displaying her body. While Ryuko undress, Barazo, Mataro, and Guts open the door to sneak a peek at Ryuko's bare chest. Before going to sleep, Mako notices Senketsu has goosebumps all over it.

Next day, Satsuki challenges Ryuko, and she reveals her Junketsu. As the uniform feeds on Satsuki's blood, her uniform squeezes on Satsuki's body, revealing ample skin. Senketsu warns Ryuko about her enemy's strength. The two warriors clash their swords. Ryuko loses some blood in the process. Satsuki easily blocks Ryuko before giving her a light tap that sends Ryuko flying. As the girls rampage throughout the school, Senketsu warns Ryuko that her blood level is dangerously low. After Satsuki blasts Ryuko, Senketsu deactivates. Satsuki claims all her actions are pure before dealing the final blow. Mako jumps in and claims that Ryuko's breasts are bigger than Satsuki's. Mako shares that her family had mentioned Ryuko had a nice rack, and she urges Ryuko to strip naked.

When Satsuki slams her blade, Ryuko blocks it with her Scissor Blade. Ryuko uses her Seki Tekko to become one with Senketsu. She regains her second wind and manages to overwhelm Satsuki. Ryuko has her Scissor Blade enter Decapitation Mode in an attempt to strip Satsuki of her uniform. Satsuki dodges in time. The girls exchange blows that shake the arena violently. Ryuko vows that she will crush everyone to make Satsuki talk. Satsuki withdraws her blade and accepts Ryuko's challenge.
